School of Science Division of Life Science 27 Cloning and Characterization of Novel Cytoskeletal Regulators Supervisor: QI Robert Zhong / LIFS Student: TSE Kai Lee / BICH Course: UROP1100, Fall UROP2100, Spring Microtubules are the cytoskeleton in eukaryotic cells that consists of α- and ß-tubulin heterodimers. They are highly conserved among species and have noticeable inherent dynamic properties. Random point mutation of human cells using CRISPR technology followed by screening could be used to discover new regulator proteins of microtubules nucleation. In this experiment, Fluorescence-Activated Cell Sorting (FACS) is used to carry out microtubule regrowth assay in mutated retinal pigmental epithelial cells (RPE-1). This report continued the UROP1100 project, which focuses on optimization of microtubule regrowth assay in suspension cells before passing through FACs, aiming to minimize other factors that may affect the signal strength. TX-100 and Tween-20 are used as detergents to permeabilize the cell. Methanol fixation was introduced and tested individually or in combination of glutaraldehyde fixation to study their effect on cell membrane permeabilization. Many literature reviews and studies propose that the presence of cholinergic system also play a crucial role in nonneural functions in human epidermis, by regulating essential activities in keratinocytes and melanocytes. The expression of cholinergic proteins, the release of acetylcholine, and the regulation of Alpha7 Nicotinic Acetylchonine Receptor (α7 nAChR) have been determined by many studies; however, the mechanism and the influence of cholinergic system on the dermatological functions have not been clearly identified. A more comprehensive understanding on the physiology and mechanism of cholinergic system would provide a more elucidated explanation, prompting a further development on therapeutic solutions and novel treatments for various diseases.
